测定意义:FAS是脂肪酸合成关键酶,催化乙酰辅酶A和丙二酰辅酶A而生成长链脂肪酸。FAS普遍表达于各种组织细胞中,在哺乳动物肝、肾、脑、肺和乳腺以及脂肪组织中表达丰富。
测定原理:FAS催化乙酰CoA、丙二酰CoA和NADPH生成长链脂肪酸和NADP+;NADPH在340nm有吸收峰,而NADP+没有;通过测定340nm 光吸收下降速率,计算FAS活性。
需要的仪器,耗材:研钵、冰、台式离心机、紫外分光光度计/酶标仪、微量石英比色皿/96孔板、可调式移液枪和蒸馏水。
